The Wuhan Institute of Virology of the Chinese Academy of Sciences has made new progress in the study of human cytomegalovirus nucleocapsid nucleation. It is found that the host cell protein WDR5 is important in the process of human cytomegalovirus nucleocapsid nucleation by assisting the formation of a nuclear complex. effect.

Relevant research results have recently been published online in the international authoritative journal "Virology Journal". This study reveals for the first time the role and mechanism of WDR5 in the human cytomegalovirus replication cycle, deepens the understanding of the replication process of this important pathogen virus, and provides new ideas for the development of antiviral drugs.

Human cytomegalovirus is a virus with a very high infection rate in the human population. Congenital human cytomegalovirus infection is the most common infectious cause of neonatal birth defects, which can lead to abnormal neurodevelopment of the fetus. The activation of latent infection often causes fatal infection in transplant recipients.

Researcher Luo Minhua introduced that WDR5 belongs to the family of WD40 repeat proteins and plays an important role in the regulation of various important physiological processes of cells. The role of this protein in the process of human cytomegalovirus infection is unclear.

The study found that human cytomegalovirus infection of fibroblasts upregulates protein levels by inhibiting the ubiquitination pathway of the host protein WDR5. Intensive studies have found that overexpression of WDR5 only slightly upregulates the production of human cytomegalovirus virions. Knockdown of WDR5 had no significant effect on virus entry into host cells, viral genome replication, viral gene expression, and nucleocapsid formation, but significantly inhibited the production of infectious virions.

Experts have conducted in-depth studies on the late steps of human cytomegalovirus replication and found that knocking down WDR5 impairs the formation of human cytomegalovirus nuclear complex, and reconstituting the expression level of WDR5 restores the production of human cytomegalovirus-infected virions. .
